JAKURA (Tofacitinib) is a targeted oral therapy for adults with active rheumatoid arthritis requiring advanced disease control. It helps regulate immune-driven inflammation and supports flexible use based on clinical evaluation.

Mechanism of Action (MoA)

Tofacitinib is a Janus kinase (JAK) inhibitor that modulates cytokine-mediated immune activation involved in RA inflammatory pathways by inhibiting intracellular signalling.

Indications

Indicated for treatment of adult patients with moderately to severely active rheumatoid arthritis with inadequate response or intolerance to methotrexate. May be used as monotherapy or with methotrexate/other nonbiologic DMARDs.

ADR (Adverse Drug Reactions)

Most frequently reported ADRs include:

• Infections (e.g., nasopharyngitis/URTI).
• Headache.
• Gastrointestinal symptoms.
• Other ADRs include laboratory abnormalities and lipid changes.

Drug Interactions

• Dose modification may be required with strong CYP3A4 inhibitors (and combined moderate CYP3A4 + strong CYP2C19 inhibitors).
• Co-administration with strong CYP inducers may reduce exposure; PI guidance to be followed.

• Hypersensitivity to tofacitinib or excipients.
